特異性及び抗原分布
BAP1(BRCA1 associated protein 1)と特異的に反応する。BAP1は、3番染色体短腕(3p21.1)上のBAP1遺伝子によりコードされる核に局在する脱ユビキチン化酵素で、転写、クロマチンリモデリング、細胞周期制御、DNA修復/組換えなどに関与している(1)~(4)(6)~(8)。生殖細胞系列におけるBAP1変異は、悪性中皮腫やブドウ膜悪性黒色腫など様々な悪性腫瘍の素因になるとされているため、BAP1はがん抑制遺伝子としても報告されている(2)(6)(7)。正常では、中皮細胞、肺胞上皮細胞(6)や血管内皮細胞、線維芽細胞、炎症性細胞(6)~(8)などの核に反応がみられる。腫瘍では、肺癌、卵巣癌などで反応がみられる(6)。悪性中皮腫(66％(6))では、免疫組織化学染色でBAP1の核からの消失(BAP1 loss)がみられるが、反応性中皮過形成ではBAP1 lossがみられないことから、悪性中皮腫と反応性中皮過形成の判別において有用であると報告されている(6)。
悪性中皮腫の組織型では、上皮型(70%)/二相型(60%)が、肉腫型(13%)/線維形成型(20%)と比較して、BAP1 lossが高率であると報告されている(6)。ブドウ膜悪性黒色腫(47%)(4)、肝内胆管癌(26%)(7)、淡明細胞型腎細胞癌(10％)(5)などの悪性腫瘍でも、BAP1 lossがみられる。
注1: BAP1が発現している細胞では、核の他に細胞質に染色がみられる場合がある。
注2: BAP1 lossを示す細胞では、核の染まりは消失しているが細胞質に染色がみられる場合がある。
